Caffeine generally serves a key ingredient in stimulant-based fat-burners because of its ability to release Noradrenaline, which induces lipolysis (fat-breakdown). Although this mechanism can certainly burn fat in the short-term, prolonged Caffeine consumption (by itself) generally results in tolerance build-up so the effects become less potent over time. This was demonstrated in a 1992 study in which 24 weeks of Caffeine intake (200mg/day) failed to induce weight-loss in humans. For Caffeine to be an effective fat-loss agent, it generally must be combined with other stimulants or synergistic compounds.

Muscle Elements lists the amount of Caffeine in one serving of 212 at 175mg, not an overwhelming dose by itself, but certainly enough to potentiate the fat-burning carried out by Synephrine and Yohimbine.


Synephrine is a naturally occurring alkaloid in such plants as Bitter Orange, although in the context of 212, it may be of synthetic origin. Either way, Synephrine has some pretty clear implications for weight-loss, specifically as a beta-adrenergic receptor agonist.

A 2011 study, published in the “International Journal of Medicinal Sciences”, found that supplementation of 50mg Syneprhine increased the metabolic rate in human subjects without affecting blood pressure or heart rate.

212 contains 30mg of Synephrine per serving, a sizable yet safe dose capable of inducing some fat-loss, especially in combination with Caffeine and Yohimbine.

Yohimbine acts as an alpha-2 receptor antagonist, meaning it inhibits the receptor responsible for blocking lipolysis. By blocking the action of this receptor Yohimbine allows for more lipolysis to occur than would normally be permitted by the alpha-adrenergic receptors.

A 2006 study, published in “Research in Sports Medicine”, found that supplementation with 20mg daily (2 10mg doses) induced significant fat-loss in athletes (Soccer players) over a two week period.

Given its role as an alpha-receptor antagonist, Yohimbine may be synergistic with compounds like Synephrine (beta-receptor agonist), and certainly has the potential to drastically increase the fat-burning effects of 212. Muscle Elements lists the amount of Yohimbine HCl at 5mg, a low enough dose to avoid some negative side effects but high enough to have an impact.

Vinpocetine is a synthetic derivative of Vincamine, extracted from Periwinkle, which has been demonstrated to increase blood flow, particularly cerebral blood flow. Vinpocetine’s potential for vasodilation has been known for quite some time, with a 1980 study from the “British Journal of Clinical Pharmacology” noting a roughly 7% increase in cerebral blood flow following infusions in healthy human subjects.

Vinpocetine has no known fat-burning capacity, but we’ve come across claims that because it increases blood flow, it increases of fatty acid oxidation. To be clear, there is absolutely no evidence to back this claim, just a tenuous (theoretical) mechanism of action. Muscle Elements does not explain the inclusion of Vinpocetine, but in the context of a fat-burner such as 212, it may function as a “support” ingredient to offset some of the negative side effects associated with certain stimulants (specifically headaches). Vinpocetine may also help maintain cognitive function during calorie restricted diets.

212 contains 5mg of Vinpocetine, not exactly a clinical dose, but still potentially beneficial.

Piper nigrum, also known as Black Pepper, contains Piperine. Several studies have found that black pepper extract, when combined with other supplements, has increased the absorption of those supplements (as measured by plasma levels). Piperine’s ability to increase absorption of other compounds is due to the inhibition of certain enzymes which breakdown most compounds, as well as the slowing of intestinal transit (increasing the amount of time these compounds are exposed to the possibility of uptake).


Evodiamine is a plant extract which appears to mimic the thermogenic effects of Capsaicin in rats. However, no human studies have been published at this time testing the effects of the extract on humans. Due to the lack of human studies available, we cannot determine with any considerable degree of certainty, the efficacy of Evodiamine.


Octopamine is a metabolite of Synephrine that is alleged to have similar fat-burner capabilities. In vitro, Octopamine has demonstrated a distinct affinity for beta-3 receptors, as opposed to the other two types. Unfortunately, since beta-3 receptors mediate fat-loss in human white adipose tissue to a lesser degree than the other beta-receptors, Octopamine may be less potent than Synephrine in a practical setting. Given other, more potent fat-burning ingredients, such as Yohimbine, being also present in the 212 formula, we wouldn’t consider Octopamine the main attraction, but it may encourage some addition fat-burning. Muscle Elements does not disclose the exact dose of Octopamine.


Taraxacum Officinale, also known as Dandelion, has a long history of use in alternative medicine as a diuretic. A 1993 study published in “Pharmaceutical Biology” pointed to the high potassium content as a possible reason for the diuretic of effect, though various compounds have been isolated and alleged to contribute to this effect.

A 2009 study published in The Journal of Alternative and Complementary Medicine found that supplementation with Dandelion Extract caused more frequent urination in subjects, but a specific mechanism of action was not identified.

Ultimately, while water certainly contributes to body weight, dehydration is not an effective long-term weight-loss strategy. Diuretics may slightly improve definition in some individuals, but fat should always be the primary target.


Uva Ursi Leaf is used in folk medicine for the treatment of urinary tract infections. The active chemical compound in the plant is a glycoside known as Arbutin, which has diuretic as well as astringent properties.

As with Dandelion, Uva Ursi will not burn any fat. It may improve definition in the short term, but is not an effective long-term weight-loss supplement.


Muscle Elements has done a pretty good job with 212, which contains a concise, properly-dosed blend of some potent stimulants. The formula also features a diutertic complex, but as our avid readers know, we generally advise against relying on diuretics for weight-loss, so in the context of 212 we’ll consider them just a little something extra. At around 50 cents per servings, 212 is competitively priced, making two servings daily a viable option for shedding a few pounds of mostly fat.
