Theacrine is an alkaloid found almost exclusively in Camellia Assamica, also known as Kucha tea. In terms of its chemical structure, Theacrine (1,3,7,9-tetramethyluric acid) is very similar to Caffeine (1,3,7-trimethylxanthine), so its physiological effects are alleged to be similar as well.

We discuss the research behind Theacrine in this article but, to make a long story short, it appears to increase perceived energy levels in humans.

Although Axis Labs does not disclose the precise dose of Theacrine present in Synthalean, it doesn’t take much to get the job done, so there isn’t any danger of under-dosing here.


Caffeine triggers the release of Noradrenaline, a potent activator of lipolysis (fat-breakdown).  This is primarily why Caffeine is a major ingredient.  In the case of Synthalean, it certainly serves as a key driver of energy/fat-burning, but the formula is by no means dependent on Caffeine alone.

Axis Labs does not disclose the amount of Caffeine in Synthalean.


Tyrosine is a non-essential amino acid (the body can produce it from Phenylalanine) which serves a precursor to Dopamine (by first being converted into L-Dopa) and Noradrenaline.

Because of this relationship, Tyrosine is alleged to increase levels of these neurotransmitters, which would theoretically lead to performance enhancement. However, research has demonstrated thatTyrosine cannot outright raise Dopamine or Noradrenaline levels upon ingestion, though it can help maintain optimal levels when depletion might otherwise occur.

Upon ingestion, Tyrosine forms substrate pool, which can then be drawn from when an acute stressor (exercise, cold exposure, etc.) causes a temporary depletion of Dopamine/Noradrenaline. For this reason, Tyrosine can be useful for maintaining cognitive function during stressful activity.

In the context of Synthalean, NALT simply serves as a means of replenishing Noradrenaline stores.


Advantra Z is a patented form of Bitter Orange Extract which is standardized for Synephrine, although Blackstone Labs doesn’t disclose the total amount of Synephrine in Cobra 6P Extreme.

Synephrine acts as a beta-receptor agonist, directly inducing lipolysis and allowing for more fat-burning than would otherwise normally occur.

For those interested, we discuss Synephrine more in-depth here, but to make a long story short:  Synephrine can be an effective fat-burner, especially when combined with other stimulants.


With the overwhelming amount of evidence indicating that Theanine enhances the positive effects of Caffeine AND reduces the negative effects, it amazes us that more brands don’t use it. Theanine and Caffeine is one of the few combinations that can actually be considered synergistic, rather than merely additive.

Theanine is effective in doses of 50-200mg and the synergistic benefits depend on how much Caffeine is being consumed with it.

Axis Labs doesn’t tell us exactly how much Theanine is present in Synthalean, but we’d estimate around 100mg.


White Willow Bark contains Salicin which, upon ingestion, is metabolized into Salicylic Acid, the same compound behind the analgesic/anti-inflammatory properties of Aspirin.

While Salicin has no clear implications for weight-loss on its own, it has been shown to enhance the effects of Ephedrine (and Ephedrine-like compounds). The mechanism of action here is via inhibition of prostaglandins, hormone-like molecules which tend to normalize the metabolic effects of beta-agonists like Ephedrine, rendering them less effective.

Salicin/Salicylic Acid was shown to potentiate the effects of Ephedrine (in mice) in a 1987 study, published in “The American Journal of Clinical Nutrition”, as well as a 1993 study from the “International Journal of Obesity and Related Metabolic Disorders”.

It’s not clear how much Salicin is present in Synthalean.


N-Methyl-Tyramine (NMT) is a Tyramine which, as mentioned above, can inhibit the reuptake of monoamines (specifically Noradrenaline), thereby amplifying and/or extending the effects of Noradrenaline releasing stimulants such as Caffeine.

Although the fat-burning potential of NMT remains unclear, it can enhance mood and therefore makes a fine addition to the Synthalean formula.


Piper nigrum, also known as Black Pepper, contains Piperine. Several studies have found that black pepper extract, when combined with other supplements, has increased the absorption of those supplements (as measured by plasma levels).

Piperine’s ability to increase absorption of other compounds is due to the inhibition of certain enzymes which breakdown most compounds, as well as the slowing of intestinal transit (increasing the amount of time these compounds are exposed to the possibility of uptake).


Synthalean is one of the more well-rounded stim-based fat-burners we’ve seen recently and is worth a shot for anyone looking for an energy/mood boosting fat-burner that actually works.  That said, this is definitely not one for stimulant-sensitive individuals.  If you don’t react well to Caffeine, you definitely won’t react well to Synthalean.
