COLEUS FORSKOHLII (10% FORSKOLIN):
Coleus Forskohlii is an Ayurvedic herb which has been studied (though not extensively) for its potential as a weight-loss agent/testosterone booster. The active component, Forskolin, has been demonstrated to increase Cyclic Adenosine Monophosphate (cAMP), the result of which is an increase in the rate of fat-loss.
A 2005 study, published in the “Journal of the International Society of Sports Nutrition”, found that 25mg Forskolin twice a day for 12 weeks was able to prevent weight gain compared to the control group in overweight women.
Another 2005 study, the subjects of which were overweight men, found that the same dose (25mg) was able to favorably influence body composition (i.e. less fat, more lean mass), which corresponded with an increase in testosterone over 12 weeks.
A more recent 2011 study noted a roughly 2.5% decrease in BMI after 2 months of supplementation. Ultimately, Forskolin may certainly result in weight-loss or at the very least, prevent weight gain. These effects may be more apparent in men, as an increase in cAMP also increases testosterone, which inherently burns fat and increases lean muscle mass.
Core Burn contains 9-10mg of Forskolin per 3 capsule serving, meaning even at two servings, the amount is slightly less than the clinically studied 25mg dose.
GREEN COFFEE EXTRACT (50% CHLOROGENIC ACID):
Green Coffee Bean Extract is generally standardized for Chlorogenic Acid content, and in the case of On Point, it has been standardized to 50%. Chlorogenic Acid is the bioactive compound primarily responsible for the moderate weight-loss effects of Green Coffee observed in multiple studies.
A 2007 study, published in the “Journal of International Medical Research”, found that 12 weeks of Green Coffee (450-500mg Clorogenic Acid) supplementation resulted in a reduction (6.9%) in glucose absorption in healthy volunteers. Researchers also noted average weight loss of 5.4 kg (almost 12 lbs) over the duration of the study in the group receiving the Green Coffee Extract.
A 2006 study, this time using a smaller dose of Green Coffee Extract (yielding 140mg Chlorogenic Acid), found no such weight-loss benefit over a 12 week period. The obvious difference between these two studies is that the dose of the first (positive) study was about 3 times the dose used in the second (negative) study.
A 2012 study found that adults who consumed GCE (containing about 315mg Chlorogenic Acid) daily lost an average of 8kg with the average reduction in body fat being about 4%.
Though Green Coffee Extract has shown mixed results when looking at all of the research as a whole, higher doses have consistently resulted in weight loss. Around 300mg of Chlorogenic Acid daily is considered an effective dose. Core Burn contains 400mg of Green Coffee Extract per three capsule serving, yielding about 200mg of Chlorogenic Acid. At two servings daily, this can be a highly effective dose.
GREEN TEA EXTRACT (90% POLYPHENOLS, 60% EGCG):
Like Green Coffee, Green Tea Extract has demonstrated the capacity for weight-loss in multiple studies, although through a very different mechanism of action. The primary compound present in Green Tea Extract, responsible for the weight-loss benefits, is Epigallocatechin gallate (EGCG).
A 2009 study, published in “The Journal of Nutrition”, found that subjects consuming 625mg Green Tea Catechins (EGCG) alongside 40mg Caffeine paired with exercise lost an average of 2.2kg (4.8lbs) compared to the subjects in the control group (consuming just Caffeine), who lost an average of 1kg (2.2lbs). These findings were corroborated by a 2009 meta-analysis, published in the “International Journal of Obesity”, which concluded that Green Tea extract tended to cause about 1.2kg (2.6lbs) reduction in bodyweight, and that effects could be amplified with Caffeine in non-caffeine tolerant individuals.
Further research has revealed that EGCG can effectively block Catechol-o-Methyl Transferase (COMT), the enzyme responsible for the degradation of Catecholamines such as Noradrenaline. The result is an indirect increase in Noradrenaline which induces lipolysis. So, while EGCG is not likely to induce noticeable weight-loss alone, when combined with Caffeine or other Noradrenaline-releasing stimulants, it can be quite synergistic. Most of the efficacy has been demonstrated using doses of 400-500mg EGCG daily, and the less caffeine-tolerant the individual, the better.
Core Burn contains 500mg of Green Tea Extract per 3 capsule serving which yields about 250mg of EGCG. At two servings daily (6 capsules total), this is a highly effective dose of EGCG.
OLEA EUROPAEA (20% OLEUROPEIN):
Olive leaf extract contains two main bioactive compounds, Hydroxytyrosol and Oleuropein, which are responsible for the majority of the adrenergic effects. Core Nutritionals discloses that this particular Olive leaf extract is standardized for 20% Oleuropein.
A 2007 study from “The Journal of Nutritional Biochemistry” demonstrated increased Noradrenaline and Adrenaline levels in rats following injection of Olive leaf extract, and this effect was primarily attributed to Oleuropein.
Similar findings were noted in a 2013 study published in “The Journal of Nutritional Biochemistry” in which rats fed a diet high in Oleuropein experienced significantly increased Noradrenaline levels (measured by urine), though it’s worth mentioning that this was not accompanied by the usual decrease in weight. In fact, Oleuropein may actually down-regulate beta-adrenergic receptors, as evidenced in a 2012 study from “The American Journal of Clinical Nutrition” (although this was in Endothelial cells, not fat-cells).
While the weight-loss implications of Olea Europaea remain unclear, it may increase Noradrenaline (pro-fat-burning neurotransmitter) which tends to increase focus and perceived energy. Core Burn contains 250mg of Oleuropein per 3 capsule serving, though given the lack of human studies this dose is difficult to interpret.
Despite the popularity of Raspberry Ketone, it has never actually demonstrated any efficacy for weight-loss in actual humans and, even in rat studies, has produced lackluster results using massive concentrations.
A 2010 in vitro study found that treatment with Raspberry Ketone increased fatty acid oxidation and lipolysis in adipocytes (fat cells). However, the amount/concentration of RK used in this study is beyond what could practically be consumed in oral supplement form.
A 2005 study, seeking to determine the weight loss effects of raspberry ketone on rats fed a high fat diet, noted dose dependent anti-obesity effects using doses of .5-4 grams/kg. This would roughly correspond to a 150lb person consuming 34-130 grams daily, a highly impractical dose.
In a 2012 study, similar effects were observed in rats, though this time with a focus on fat accumulation in the liver resulting from a high fat diet. The only human study that exists grouped Raspberry Ketone in with several other popular weight-loss ingredients so the effects cannot be attributed to raspberry ketones alone.
On a molecular level, Raspberry Ketone certainly demonstrates anti-obesity effects, but the doses used to achieve these effects are far more than what the average human could practically consume.
Core Burn contains a pretty standard 100mg of Raspberry Ketone per 3 capsule serving, but even at a much higher dose, it’s unlikely that it would contribute much to the overall effect.
Rauwolscine (also known as alpha-yohimbine) is what is known as a ‘stereoisomer’ of Yohimbine, meaning it is chemically similar in structure. Because of this similarity, Rauwolscine produces similar effects, although perhaps to a milder degree. In the context of the PX formula, Rauwolscine functions as an Alpha-Receptor Antagonist which may enhance the fat-burning induced by the other stimulants (Beta-Receptor Agonists) in the formula. For individuals who do not regularly consume Rauwolscine, it may also increase focus/intensity/perceived energy via release of noradrenaline.
Bacopa Monnieri is an herb which has traditionally been used as a nootropic and anxiolytic. However, in the context of Thermo Fire, the implied claim is that Bacopa Monnieri is able to influence Thyroid Hormones, thus positively impacting body weight.
A 2002 study from “Ethnopharmacology” found that 200mg/kg daily of Bacopa Extract was able to raise T4 (a Thryoid Hormone) by about 42% in male mice. Currently, the effects of Bacopa on Thyroid hormones have not been studied in human subjects, and it is unknown how beneficial it can be for weight loss. Logically, the most likely individuals to benefit from Bacopa supplementation would be those with Thyroid issues and abnormally low levels of T4.
Given a dose of roughly 200mg Bacopa per 3 capsule serving, Core Burn likely won’t have much of an influence on Thyroid Hormone levels, although there may be some marginal benefit.
Higenamine, also known as Norcoclaurine acts as Beta(2)Adrenergic Agonist (same mechanism as Ephedra), meaning it stimulates the Beta(2) Adrenergic Receptors which induce lipolysis (fat burning). In addition to its fat-burning potential, Higenamine has also been demonstrated in vitro to increase acetylcholine levels, though these findings have not yet been replicated in humans. Overall, there is certainly preliminary support for Higenamine as a fat-burner and potential ergogenic aid, but because no human studies exist there is recommended effective dose. Since Higenamine acts as a Beta Receptor Agonist, it may synergistically enhance the fat-burning effects of Alpha Receptor Antagonists such as Rauwolscine, though this relationship has not been the subject of any published studies thus far.
Core Burn contains 30mg of Higenamine per 3 capsule serving, slightly more than the average competitor, and an effective dose according to anecdotal reports.
Picamilon (alternatively spelled ‘Pikamilon’) is formed by combining Niacin (vitamin B3) and GABA (the primary inhibitory neurotransmitter in mammals). Pikamilon is able to effectively cross the blood-brain-barrier where it is converted into GABA. Since GABA is an inhibitory neurotransmitter (as opposed to excitatory) it may produce anxiolytic effects when levels are increased beyond normal. However, Picamilon does not produce the sedative like effects of other GABA pro-drugs and instead is commonly used as a nootropic (cognitive enhancer) because it has also been demonstrated to increase cerebral blood flow due to its Niacin component (Niacin is a vasodilator). Despite a fair amount of efficacy demonstrated in animal studies for both cerebral vasodilation and as a non-sedative anxiolytic, human studies remain scarce.
Picamilon does not necessarily have any clear weight-loss implications, but may enhance the cognitive aspect of the Core Burn formula, so we feel it still has a place.
Caffeine generally serves a key ingredient in stimulant-based fat-burners because of its ability to release Noradrenaline, which induces lipolysis (fat-breakdown). Although this mechanism can certainly burn fat in the short-term, prolonged Caffeine consumption (by itself) generally results in tolerance build-up so the effects become less potent over time. This was demonstrated in a 1992 study in which 24 weeks of Caffeine intake (200mg/day) failed to induce weight-loss in humans. For Caffeine to be an effective fat-loss agent, it must be combined with other stimulants or synergistic compounds such as EGCG.
THE BOTTOM LINE:
Core Burn contains a diverse range of fat-loss ingredients, some of which have been more extensively studied (and proven) than others. Certain individual ingredients may be slightly under-dosed, but at two servings of 3 capsules (6 capsules total) daily, Core Burn may certainly function as an effective fat-burner.
