Adipo-X is a fat-burner made by Axis Labs which contains a variety of weight-loss ingredients, some stimulant and some non-stimulant…[Skip to the Bottom Line]
Caffeine generally serves a key ingredient in stimulant-based fat-burners because of it’s ability to release Catecholamines (Noradrenaline, Dopamine, etc.), which induce lipolysis (fat-breakdown). Although this mechanism can certainly burn fat in the short-term, prolonged Caffeine consumption (by itself) generally results in tolerance build-up so the effects become less potent over time. This was demonstrated in a 1992 study in which 24 weeks of Caffeine intake (200mg/day) failed to induce weight-loss in humans. For this reason, for Caffeine to be an effective fat-loss agent, it must be combined with other stimulants such as Synephrine.
Axis Labs does not disclose the exact dose of Caffeine present in the Adipo-X formula, but we’d estimate it contains anywhere between 200-300mg per serving which, combined with Synephrine, would be enough to kick-start the fat-burning process in the average (non-caffeine tolerant) individual.
Tyrosine is a non-essential amino acid (the body can produce it from Phenylalanine) which serves a precursor to Dopamine (by first being converted into L-Dopa) and Noradrenaline. Because of this relationship, Tyrosine is alleged to increase levels of these neurotransmitters, which would theoretically lead to performance enhancement. However, research has demonstrated that Tyrosine cannot outright raise Dopamine or Noradrenaline levels upon ingestion, though it can help maintain optimal levels when depletion might otherwise occur.
Upon ingestion, Tyrosine forms substrate pool, which can then be drawn from when an acute stressor (exercise, cold exposure, etc.) causes a temporary depletion of Dopamine/Noradrenaline. For this reason, Tyrosine can be useful for maintaining cognitive function during long-term, mentally strenuous exercise.
Adipo-X contains L-Tyrosine AKG, which is formed by combining Tyrosine with alpha-Ketoglutaric Acid, and is said to be better absorbed than standard L-Tyrosine. Whether this is actually true or not is unclear, as no studies have actually confirmed this alleged absorption enhancement. Regardless of whether it is better absorbed, Tyrosine (in any form) doesn’t have any direct weight-loss implications, but may help to maintain levels of the fat-burning Catecholamine neurotransmitters.
Despite the popularity of Raspberry Ketone, it has never actually demonstrated any efficacy for weight-loss in actual humans and, even in rat studies, has produced lackluster results using massive concentrations.
A 2010 in vitro study found that treatment with Raspberry Ketone increased fatty acid oxidation and lipolysis in adipocytes (fat cells). However, the amount/concentration of RK used in this study is beyond what could practically be consumed in oral supplement form.
A 2005 study, seeking to determine the weight loss effects of raspberry ketone on rats fed a high fat diet, noted dose dependent anti-obesity effects using doses of .5-4 grams/kg. This would roughly correspond to a 150lb person consuming 34-130 grams daily, a highly impractical dose.
In a 2012 study, similar effects were observed in rats, though this time with a focus on fat accumulation in the liver resulting from a high fat diet. The only human study that exists grouped Raspberry Ketone in with several other popular weight-loss ingredients so the effects cannot be attributed to raspberry ketones alone.
On a molecular level, Raspberry Ketone certainly demonstrates anti-obesity effects, but the doses used to achieve these effects are far more than what the average human could practically consume. There are other substances with much more pronounced fat-burning capabilities than raspberry ketone when compared by weight.
Axis Labs touts Raspberry Ketone as a revolutionary ingredient which, unfortunately, sort of detracts from the validity of the claims regarding the entire product. Raspberry Ketone, while once thought to be a revolutionary fat-burning ingredient, has fallen far short of most expectation when studied directly and there is no evidence to suggest any meaningful weight-loss benefits can be obtained through oral consumption.
Advantra Z is a patented form of Bitter Orange Extract which is generally standardized for Synephrine (in this case 30%), which has some pretty clear weight-loss implications, assuming the right dosage.
A 2011 study, published in the “International Journal of Medicinal Sciences”, found that supplementation of 50mg Syneprhine increased the metabolic rate in human subjects without affecting blood pressure or heart rate.
Similarly to Ephedrine, Synephrine is a beta-receptor agonist and an alpha-receptor antagonist, the net effect of which is an increase in lipolysis. Unfortunately, Axis Labs does not give us any indication of how much Synephrine is actually present in the Adipo-X formula, but considering Synephrine is effective at pretty low doses, there is no obvious flaw that we can see (based on the proprietary blend as a whole).
Ginger is a very common household digestive aid, commonly prescribed by loving mothers for stomach problems. However, a 2012 pilot study from “Metabolism” found that 2 grams of Ginger consumed with a meal increased caloric expenditure for several hours following the meal in overweight men. Ginger has also been shown to increase Insulin secretion via acting as a serotonin receptor antagonist (serotonin normally suppresses insulin to some degree) in mice, though a human study using 1 gram of Ginger failed to replicate this results.
Of course, Adipo-X doesn’t contain anywhere near the above-mentioned doses (1-2g), but it is possible that, at whatever dose is present in the formula, Ginger is still somewhat effective.
Paprika, yes the stuff you sprinkle on deviled eggs, is believed to be somewhat similar to Cayenne, with regards to fat-burning. Given that it contains a number of pungent alkaloids, this is believable, but at this time there are no studies testing any isolated alkaloids, or even Paprika as a whole, for weight-loss benefit. In the context of Adipo-X, Paprika doesn’t hurt, but we definitely wouldn’t consider it a “key ingredient” or anything.
Capsicum Annum, more commonly known as Cayenne Pepper, is generally standardized for its active component, Capsaicin, which has thermogenic properties and clear implications for weight-loss in humans.
A 1997 study, the subjects of which were rats, found that Capsaicin supplementation was able to increase adrenaline-induced fatty acid utilization, which increased exercise (swimming) endurance.
A more recent (2007) study noted an increase in fat oxidation (relative to placebo) during low intensity exercise in healthy adult males who consumed 150mg of capsaicin one hour before exercise.
Due to the lack of disclosure, the dose of Capsaicin found in Adipo-X is unclear.
Coleus Forskohlii is generally standardized for the active compound Forskohlin, which has been demonstrated to increase Cyclic Adenosine Monophosphate (cAMP), the result of which is an increase in the rate of fat-loss.
A 2005 study, published in the “Journal of the International Society of Sports Nutrition”, found that 50mg daily (two 25mg doses) for 12 weeks was able to prevent weight gain compared to the control group in overweight women. Another 2005 study found that the same dose (25mg twice daily) was able to favorably influence body composition (i.e. less fat, more lean mass), which corresponded with an increase in Testosterone over 12 weeks. A more recent 2011 study noted a roughly 2.5% decrease in BMI after 2 months of supplementation.
Forskohlin is one of the more effective non-stimulant fat-burners on its own, and can potentially amplify the effects of other fat-burning compounds. Although Axis Labs does not directly disclose the dose of Forskohlin present in the Adipo-X formula, the particular form of Coleus Forskohli is standardized for 40% Forskohlin, further increasing the odds that there is an effective dose present.
GREEN TEA EXTRACT:
Multiple studies have confirmed Green Tea Extract can influence fat-loss to a statistically significant degree. Although this effect was originally thought to be related to caffeine content, more recent research has pointed to a green tea catechin known as Epigallocatechin gallate (EGCG) as the compound primarily responsible for these effects.
A 2009 study, published in “The Journal of Nutrition”, found that subjects consuming 625mg Green Tea Catechins (EGCG) alongside 40mg Caffeine paired with exercise lost an average of 2.2kg (4.8lbs) compared to the subjects in the control group (consuming just Caffeine), who lost an average of 1kg (2.2lbs). These findings were corroborated by a 2009 meta-analysis, published in the “International Journal of Obesity”, which concluded that Green Tea extract tended to cause about 1.2kg (2.6lbs) reduction in bodyweight, and that effects could be amplified with Caffeine in non-caffeine tolerant individuals.
Further research has revealed that EGCG can effectively block Catechol-o-Methyl Transferase (COMT), the enzyme responsible for the degradation of Catcholamines such as Noradrenaline. The result is an indirect increase in Noradrenaline which induces lipolysis. So, while EGCG is not likely to induce noticeable weight-loss alone, when combined with Caffeine or other Noradrenaline-releasing stimulants, it can be quite synergistic. Most of the efficacy has been demonstrated using doses of 400-500mg EGCG daily, and the less caffeine-tolerant the individual, the better.
Green Tea is perhaps the only ingredient in Adipo-X that is quite obviously under-dosed, based on its position in the proprietary blend. Considering that Axis Labs uses a form of Green Tea Extract that is 45% EGCG, there would have to be about 700mg to yield a truly effective dose in a single serving of Adipo-X.
Vinpocetine is a synthetic derivative of Vincamine, extracted from Periwinkle, which has demonstrated the ability to increase blood flow, particularly cerebral blood flow. Vinpocetine’s potential for vasodilation has been known for quite some time, with a 1980 study from the “British Journal of Clinical Pharmacology” noting a roughly 7% increase in cerebral blood flow following infusions of Vinpocetine in healthy human subjects.
While claims regarding the implications of Vinpocetine for weight-loss tend to vary from product to product, it does have an indirect way of improving stimulant-based fat-burners. Because Vinpocetine is truly an effective cerebral vasodilator, it can reduce stimulant-induced migraines and potentially enhance cognitive ability, as evidenced in a 1985 study, published in the “European Journal of Clinical Pharmacology”, which noted decreased reaction time in subjects taking 40mg of Vinpocetine.
Adipo-X probably contains nowhere near the 40mg dose used in the above-mentioned study, but may still contain a mildly effective dose.
PIPER NIGRUM FRUIT EXTRACT (BIOPERINE):
Piper nigrum, also known as Black Pepper, contains Piperine. Several studies have found that black pepper extract, when combined with other supplements, has increased the absorption of those supplements (as measured by plasma levels). Piperine’s ability to increase absorption of other compounds is due to the inhibition of certain enzymes which breakdown most compounds, as well as the slowing of intestinal transit (increasing the amount of time these compounds are exposed to the possibility of uptake).
THE BOTTOM LINE:
Adipo-X contains a wide array of fat-burning ingredients, some of which are more effective than others. The bulk of the weight-loss potential comes from the the combination of Caffeine, Synephrine, Forskolin, and Capsaicin, with the remaining of the ingredients playing more of a “support role”. At about 50 cents per dose, Adipo-X is on the inexpensive side, compared to competing products, and at 1-2 daily doses, may help to burn of some excess body fat.
